Arabic Abstract
يُعد البنزو[a]بيرين والكريسين من الهيدروكربونات العطرية عديدة الحلقات (PAHs) ذات الوزن الجزيئي العالي، المعروفة بثباتها البيئي وسمّيتها، إذ تُعتبر من أكثر المركّبات مقاومةً للتحلّل ومسببةً للسرطان. تهدف هذه الدراسة إلى بحث قابلية هذه المركّبات للتحلّل الحيوي باستخدام سلالتين بكتيريتين هما أوكروبكتروم إنتيرميديام بي سي 1 وكوبرفايدوس تايوانينسِس إل إيه، والمعزولتين من رواسب ساحلية في المملكة العربية السعودية. أُجريت التجارب في وسط بوشنل–هاس المعدني )درجة حموضة 7، ودرجة حرارة 37 م (° لمدة 18 يوماً، مع إضافة كل مركّب من مركّبات PAH بتركيز 50 جزءاً في المليون كمصدر وحيد للكربون. تمّ تتبّع عملية التحلّل باستخدام تقنية الكروماتوغرافيا الغازية المقترنة بمطياف الكتلة (GC–MS)، في حين تمت متابعة نمو البكتيريا طيفياً. أظهرت كلتا السلالتين قدرة عالية على تحلّل هذه المركّبات المقاومة، حيث حققت السلالة إل إيه أعلى كفاءة )إزالة % 99 ≈ للكريسين و≈95 %للبنزو[a]بيرين(، تلتها السلالة بي سي 1 بفارق بسيط. كما ظهر ناتج تحلّل مؤقت ذو أربع حلقات (m/z 228) أثناء تحلّل البنزو[a]بيرين، مما يشير إلى تحوّله إلى مركّب وسيط مشابه للكريسين قبل تفككه الكامل. لم يؤدّ استخدام المزرعة المشتركة للسلالتين إلى تحسين الكفاءة الإجمالية. تُظهر هذه النتائج القدرة الإنزيمية للبكتيريا المحلية السعودية على تحلّل الهيدروكربونات العطرية المسرطنة دون الحاجة إلى ركائز مساعدة خارجية، وتبرز دورها المحتمل في المعالجة الحيوية المستدامة للبيئات الملوثة بالنفط في المناطق الجافة
English Abstract
Benzo[a]pyrene and chrysene are high-molecular-weight polycyclic aromatic hydrocarbons (PAHs) known for their environmental persistence and toxicity, as they are among the most recalcitrant and carcinogenic PAHs. This study investigated the biodegradability of those compounds using bacterial strains Ochrobactrum intermedium BC1 and Cupriavidus taiwanensis LA isolated from Saudi Arabian coastal sediments. Experiments were conducted in Bushnell–Haas mineral medium (pH 7, 37 °C) for 18 days, with each PAH added at 50 ppm as the sole carbon source. The degradation process was monitored using gas chromatography–mass spectrometry (GC–MS), while bacterial growth was followed spectrophotometrically. Both strains demonstrated a strong ability to degrade these recalcitrant compounds, with LA achieving the highest removal efficiency (≈ 99% for chrysene and ≈ 96% for BaP), followed closely by BC1. A transient four-ring metabolite (m/z 228) appeared during BaP degradation, suggesting conversion to a CHR-like intermediate prior to complete breakdown. The co-culture of both strains did not enhance the overall efficiency. These findings demonstrate the enzymatic capacity of indigenous Saudi bacteria to degrade carcinogenic PAHs without external co-substrates and highlight their potential role in sustainable bioremediation of petroleum-contaminated environments in arid regions.
