Arabic Abstract
تستلزم المشكلة المتفاقمة لمقاومة مضادات الميكروبات تطوير منصات قاتلة للبكتيريا غير تقليدية، قادرة على تفكيك البكتيريا من خلال آليات متعددة تعمل في الوقت نفسه. وقد قيّم هذا البحث النشاط المضاد للميكروبات لمركبات الفضة–الأنابيب الكربونية النانوية (Ag-CNT) المُحضَّرة في جامعة الملك فهد للبترول والمعادن، كما حدّد التأثير الإضافي الناتج عن دمج هذه المركبات مع المضاد الحيوي السيبروفلوكساسين. وافترضت الدراسة أن هذا الفعل المشترك يستفيد من الخصائص الفيزيائية المسببة لاضطراب الغشاء التي تمتلكها الأنابيب الكربونية النانوية متعددة الجدران (MWCNTs)، ومن النشاط المضاد للميكروبات للفضة عبر آلياتها الأيونية والأكسدية، إلى جانب النشاط المضاد للبكتيريا واسع الطيف للسيبروفلوكساسين. وعلى وجه التحديد، جرى تحضير الأنابيب الكربونية النانوية متعددة الجدران المحمّلة بجسيمات الفضة النانوية (Ag-CNT) عن طريق الاختزال الموضعي لأيونات الفضة (+Ag) على أسطح الأنابيب الكربونية النانوية متعددة الجدران المُفعَّلة حمضيًا، ثم وُصفت هذه المواد للتأكد من نجاح تكوينها وملاءمتها البنيوية للتقييم المضاد للبكتيريا. وأكد كلٌّ من تحليل FTIR وتحليل XRD، على التوالي، وجود تفاعل بين أنواع الفضة والبنية الوظيفية للأنابيب النانوية، مع احتفاظ المادة بطابعها الكربوني وظهور إسهامات بلورية إضافية تُعزى إلى إدماج الفضة. وتم قياس الأداء المضاد للبكتيريا ضد البكتيريا النموذجية سالبة الغرام Escherichia coli باستخدام طريقة التخفيف الكبير في المرق، من خلال تحديد التركيز المثبط الأدنى (MIC) والتركيز القاتل الأدنى للبكتيريا (MBC)، كما جرى تقييم تعزيز فعالية المضاد الحيوي من خلال دمج Ag-CNT مع السيبروفلوكساسين. وأظهرت النتائج أن Ag-CNT يمتلك نشاطًا مضادًا للبكتيريا جوهريًا وقابلًا للقياس ضد E. coli، حيث بلغ التركيز المثبط الأدنى 62.5 ميكروغرام/مل، وبلغ التركيز القاتل الأدنى للبكتيريا 250 ميكروغرام/مل، في حين أظهر السيبروفلوكساسين تركيزًا مثبطًا أدنى قدره 0.5 ميكروغرام/مل. وقد ثبّط مزيج Ag-CNT–السيبروفلوكساسين نمو البكتيريا عند تركيز 31.25 ميكروغرام/مل من Ag-CNT + 0.25 ميكروغرام/مل من السيبروفلوكساسين، مما يدل على انخفاض التراكيز التثبيطية لكلا العاملين في المعالجة المشتركة. وأظهر التفسير الإضافي باستخدام منهج التركيز التثبيطي الكسري أن التفاعل كان إضافيًا لا تآزريًا حقيقيًا. وقد دُعم ذلك أيضًا من خلال تجارب الزرع على الأطباق المُعادة وتقييم عدد المستعمرات، إذ أكدت هذه النتائج نقطة النهاية القاتلة للبكتيريا للمزيج، وأظهرت تعافيًا بكتيريًا تدريجيًا عند التراكيز الأدنى من ذلك.
English Abstract
The aggravating issue of antimicrobial resistance necessitates non-traditional bactericidal platforms that dismantle bacteria through multiple mechanisms simultaneously. This research evaluated the antimicrobial activity of the Ag-CNT composites synthesized at KFUPM and determined additive effect of combining these composites with the antibiotic, ciprofloxacin. It was hypothesized that this combined action would leverage the physical, membrane-disrupting properties of Multi-Walled Carbon Nanotubes (MWCNTs), the ionic and oxidative antimicrobial activity of silver with the broad-spectrum antimicrobial activity of ciprofloxacin. Specifically, AgNP‑loaded MWCNTs (Ag‑CNT) was synthesized by in‑situ reduction of Ag⁺ onto acid‑functionalized MWCNTs and were characterized to confirm their successful formation and structural suitability for antibacterial evaluation. FTIR analysis and XRD analysis substantiated an interaction between silver species and the functionalized nanotube structure and confirmed retained carbonaceous character with additional crystalline contributions attributable to silver incorporation respectively. Antimicrobial performance against the model Gram-negative bacterium Escherichia coli was quantified using broth macrodilution through minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) determination, and antibiotic potentiation was evaluated by combining Ag-CNT with ciprofloxacin. The results demonstrated that Ag-CNT possessed measurable intrinsic antibacterial activity against E. coli, with an MIC of 62.5 µg/mL and an MBC of 250 µg/mL, while ciprofloxacin showed an MIC of 0.5 µg/mL. The Ag-CNT-ciprofloxacin combination inhibited bacterial growth at 31.25 µg/mL Ag-CNT + 0.25 µg/mL ciprofloxacin, indicating reduced inhibitory concentrations for both agents in the combined treatment. Further interpretation using the fractional inhibitory concentration approach showed that the interaction was additive rather than truly synergistic. This was further supported by reproduced plating experiments and colony count assessment, which confirmed the bactericidal endpoint of the combination and demonstrated progressive bacterial recovery below that concentration.
