Arabic Abstract

لقد تمت دراسة السلوك الكشطي الكھروكيماوي لمكونات الحمض النووي على أسطح أقطاب الكربون المختلفة. وتم تطويرطريقة كھركيميائية بسيطة وحساسة وموثوقة من أجل الفحص والتحليل للأدينين والجوانين و الادينوزين و الجوانوزين على أسطح ھذه الاقطاب الكربونية الجديدة. ومن التقنيات الكھركيميائية المستخدمة في Square-) والفولتامترية الكشطية رباعية الموجة (Cyclic Voltammetry) الدراسة الفولتامترية الدائرية والفولتامترية ( Linear-Sweep Voltammetry) والفولتامترية المسحية الخطية (Wave Voltammetry .(Differential Pulse Voltammetry) الكشطية النابضة وقد تمت دراسة مختلف العوامل المؤثرة على تقدير تركيزات دقيقة من ھذه المركبات محل الدراسة على وكذلك الكربون (Glassy Carbon Paste) أسطح الاقطاب الكربونية الكھربائية التالية: عجينة الكربون الزجاجي .(Graphite Pencil) والكربون الجرافيتي (Glassy Carbon) الزجاجي لقد تمت أيضا دراسة وتحديد أفضل الشروط المناسبة لھذه الطرق المطورة. وكذلك امكانية استخدام ھذه الطرق لتحديد ھذه المكونات في وسط بيولوجي وكذلك دراسة تأثير وجود بعض المركبات العضوية الشائعة. وقد وأخيراً تمت بنجاح مقارنة ھذه الطرق .(ssDNA) أثبتت الطرق المطوّرة نجاحھا للتحديد المباشر للحمض النووي .(HPLC) الكھربية المطوّرة مع تقنية الكروماتوجرافيا السائلة عالية الأداء

English Abstract

The attractive stripping performance of nucleic acid constituents were characterized at different carbon composite electrode surfaces. A simple, sensitive and reliable electrochemical method was developed for the investigation and analytical determination of adenine (A), guanine (G), adenosine (AS) and guanosine (GS) at different composite carbon electrode surfaces. Different electroanalytical techniques were used as cyclic voltammetry (CV), square wave stripping voltammetry (SWV), linear sweep voltammetry (LSV) and differential pulse voltammetry (DPV). Factors influencing the trace detection of these compounds at glassy carbon paste (GCP), glassy carbon (GC) and graphite pencil (GP) composite electrode surfaces were assessed. All developed methods were optimized and the figures of merit were determined. The possibility of the detection in biological samples, interference of common organic compounds, and direct determination of ssDNA were tested successfully. Finally the developed methods were compared to HPLC.
